Description
This compound is a synthetic 37-residue acylated amylin analogue. It carries a C20 diacid (eicosanedioic acid) side chain attached through a gamma-glutamate spacer, a C-terminal amide, and an intramolecular disulfide bridge. It is produced by solid-phase peptide synthesis and supplied as a lyophilized powder. The acyl side chain confers reversible albumin binding, and it is also referred to in the literature as AM833.
Its backbone derives from amylin, a 37-residue peptide, with substitutions selected during structure-activity work to reduce the fibrillation propensity that makes the native sequence difficult to handle in solution. Cagril is classified as a non-selective agonist of the calcitonin family of class B G protein-coupled receptors. This family includes the calcitonin receptor (CTR) on its own and the amylin receptors (AMY1, AMY2, AMY3), which are heterodimers of CTR with receptor activity-modifying proteins RAMP1, RAMP2, and RAMP3.
